Reviewed by ICLAC (ref: 190616). L-02 was reported to be established from normal liver cells but its STR profile corresponds to HeLa. Samples tested in this study were directly deposited in the cell line repository that performed the investigation. L-02 has a derivative: HL-7702BaPT. No STR profiling information was available for the derivative at the time of review.

Instruction

Use exclusive Red Cotton system to design a sgRNA fragment knockout strategy for the HIF1A gene, clone the two sgRNAs into CRISPR-U™ vector, transfect the vector into the L-02 cell line by electroporation/lentivirus method, use the limited dilution method for single-cell cloning and use the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000) for nucleic acid lysis and PCR amplification on the single-cell clones, followed by Sanger sequencing to verify the genotype. Upon passing the verification, HIF1A gene knockout HIF1A Knockout cell line (L-02) will be expanded and cryopreserved.
